DESCRIPTION

Oxacillin Injection, USP is a sterile injectable product containing oxacillin which is added as oxacillin (oxacillin (oxacillin for injection) for injection) sodium, a semisynthetic penicillin derived from the penicillin nucleus, 6-aminopenicillanic acid. The chemical name of oxacillin (oxacillin (oxacillin for injection) for injection) sodium is 4-Thia-1-azabicyclo[3.2.0]heptane-2-carboxylic acid, 3,3- dimethyl-6-[[(5-methyl-3-phenyl-4- isoxazolyl)carbonyl]-amino]-7-oxo-, monosodium salt, monohydrate, [2S-(2α,5α,6β)]-. It is resistant to inactivation by the enzyme penicillinase (beta-lactamase). The molecular formula of oxacillin (oxacillin (oxacillin for injection) for injection) sodium is C19H18N3NaO5S•H2O.

INDICATION

Oxaciilin is an isoxazolyl penicillin used similarly to flucloxacillin in the treatment of infections due to staphylococci resistant to benzylpenicillin.

DOSAGE AND ADMINISTRATION

Oxacillin is given by mouth or by injection as the sodium salt. Doses are expressed in terms of the equivalent amount of oxacillin.

Usual adult oral doses are 0.5 to 1g of oxacillin every 4 to 6 hours. Similar doses may be givenby intramuscular injection, by slow intravenous injection over about 10 minutes, or by intravenous infusion.

Children weighing less than 40 kg may be given 50 to 100mg/kg daily in divided doses by mouth or parentally.

Doses may be increased in severe infections.

DRUG INTERACTIONS

Probenecid prolongs the half-life of benzylpenicillin by competing with it for renal tubular secretion and may be used therapeutically for this purpose. Benzylpenicillin may also interact with bacteriostatic anti-bacaterials such as chloramphenicol and tetracyclines, and may be incompatible in vitro with other drugs, including a number of other antibacterials.

ADVERSE EFFECTS AND PRECAUTIONS

Hepatitis and cholestatic jaundice have been reported occasionally with flucloxacillin and may be delayed in onset for up to several weeks after treatment has been stopped; older patients and those receiving flucloxacillin for more than 2 weeks are at greater risk. Agranulocytosis and neutropenia have been associated rarely with isoxazolyl penicillin such as flucloxacillin Phlebitis has followed intravenous infusion.
